xMAP Technology and Multiplex Platform

The Enzyme-Linked Immunosorbent Assay (ELISA) has long been the main instrument for detecting interest analytes in biological samples for both clinical research and diagnostics. However, this technology has some limitations: it requires a relatively large sample size and is subject to unspecific and high background phenomena. In addition, most ELISA assays are based on enzyme amplification of the signal in order to obtain a higher sensitivity.

Such amplification is not always linear and can therefore alter the results. In recent years, a new technology has emerged that offers the advantages of ELISA, but it allows for greater productivity and flexibility, using a reduced sample volume and relatively lower costs. Luminex xMAP technology is based on the use of microspheres that allow both single and multiplex dosages for qualitative and quantitative determination of nucleic acids and proteins.

Microgem Lab provides the “Analysis of Proteins with xMAP Technology and Multiplex Platform” service in circulating fluids (plasma, serum, cerebrospinal fluid, washing fluids), cellular and tissue extracts and cell culture supernatants using xMAP technology and multiplex platform. Analysis of single or multiple analytes up to a maximum of 50 analytes per reaction. Possibility to analyze biological samples of human, mouse, rat, dog, cat.

Workflow of the “Analysis of proteins with xMAP technology and multiplex platform” service

  • Consultancy – A free consultation will be provided for project optimization (number of samples to be analyzed, methodology, costs). During the consultation phase, advice will be provided on the sample preparation method to be sent.
  • Sample Requirements –
    • 5 ml of culture supernatants without debris
    • 3 ml cell lysate at protein concentration of 400-1000 ug/ml
    • 3 ml of plasma from EDTA or citrate blood samples
    • 3 ml of serum
  • Test setup – The assay will be conducted in a 96-well plate where all the reagents and sample aliquots to be analyzed will be added.
  • Data Analysis – Quantitative data analysis.
  • Final Report – At the end of the project, a final report will be provided including the results of the various stages of project development. The results presented in the report can be discussed with the scientific team.
