Real-Time PCR
Quantitative Real-Time PCR (qPCR) allows to detect and quantify target nucleic acids by means of fluorescent dyes intercalating in the double strands (SYBRGreen) of nucleic acids or specific probes to which fluorescent reports are linked. Microgem Lab offers the “qPCR Analysis” service, using SYBRGreen.

Sample Preparation
- Cells and tissues: RNA isolation, cDNA synthesis
- From RNA: cDNA synthesis
Sample Requirements
- 3 ug of total RNA, DNase I treated
- RNA samples are required to have an OD 260/280 ratio between 1.8-2.0 and 260/230 ratio ≥7
Please supply a shot of 1.5 % denaturating agarose gel.
Experimental Design and Execution
- Gene expression analysis performed in triplicate for target interest and the “housekeeping” gene
- Negative controls without templates
Data Analysis
- Analysis of expression levels related to the “housekeeping” gene
